本公司提供两套3'UTR报告载体。在pEZX-MT05载体中,我们将3'UTR序列(来源于公开的基因序列数据库)插入到Gaussia荧光素酶报告基因下游,在哺乳动物细胞中由SV40启动子驱动转录生成Gaussia荧光素酶(Gluc)和3'UTR靶标序列的嵌合mRNA,因此无需裂解细胞实时监测Gluc的表达,研究mRNA-miRNA的相互作用。在相同载体(pEZX-MT05)上带有分泌型碱性磷酸酶(SEAP)报告基因。CMV启动子驱动转录的SEAP作为同质粒内的对照,这种双报告系统可以对多样本常规转染进行精确比较。
pEZX-MT06是Firefly/Renilla双荧光素酶报告载体。3'UTR序列在Firefly(hLuc)荧光素酶报告基因下游,CMV启动子驱动转录的Renilla(hRLuc)作为同质粒内的对照。通过裂解细胞检测hLuc和hRLuc的表达。
